Unlike conventional T cells, innate-like T cells are characterized by the induction of rapid cytokine secretion and early response in infectious and inflammatory diseases. We have revealed the requirement, endocytic pathway, and the conservation of antigen presentation for the activation of mucosal-associated invariant T (MAIT) cells. Notably, the mycobacterial non-peptidic molecules are able to activate MAIT cells, which were shown to be protective against mycoabcterial infections. We also set up a functional lipidomic platform to profile the lipid metabolites associated with antigen presenting molecules CD1 proteins using mass spectrometry and study the lipid antigens for T cell activation. These studies revealed the CD1b-associated scaffold lipid diacylglycerol as an adjuvant in anti-mycobacterial immune responses and CD1a-presented headless lipids as antigens in skin immune regulation. These antigenic molecules potentially provide new tools for fighting infections and inflammations.
主讲人简介:
黄守雄,辛辛那提大学医学院环境健康系辛辛那提儿童医院免疫生物学系助理教授(Assistant Professor)。1993年北京中医药大学获学士学位,2003年俄亥俄州立大学获博士学位,随后在圣路易斯华盛顿大学医学院和哈佛大学医学院作博士后研究和讲师(Instructor)。研究兴趣包括非多肽抗原的递呈,天然T细胞(innate-like T cells)在结核病和炎症性疾病中的作用,及环境毒物对免疫功能的影响。其研究阐明了粘膜相关非多样性T细胞(Mucosal-associated invariant T cell, MAIT)的激活机制并通过建立了免疫脂质组学(Lipidomics)的平台比较研究了脂类抗原的分子结构和对T细胞激活的功能。这些发现已在Nature Immunology, J Exp Med, PNAS, J Biol Chem等杂志发表,并对临床诊断和治疗传染性和炎症性疾病有潜在的影响。
